    PS3 Media server supports MKV.

you can dual boat your PS3 to run LINUX and XBMC.

and will PS# Media Server support a UPnP device (in this case, my router)? or does it have to be off a computer?

    I use the same router.

you can also directly plug USB devices into the PS3 but I believe it needs to be put into specific folder names.

ya I don't really want to do that as I still want my other PCs to be able to access the files. I have other stuff besides media files on the drive.

AFAIK you simply need to have them within a folder on the HD under a specific path name, it won't effect the usage at all. like you're movies need to all be in a folder called "movies" or whatever the path needs to be, I stream everything so i've never actaully done it myself.

It does work though as i've watched a movie off my friends thumbdrive.
